Does Bone Marrow Aspirate Augment Bone Formation with a Hydroxyapatite Scaffold?

چکیده
Introduction: Bone graft is crucial in the repair and regeneration of large or criticalsized defects and hydroxyapatite has been widely used in many clinical conditions as a bone graft substitute material. However, in the older patient group the challenges faced by the surgeon to repair and regenerate defects with good quality bone can be difficult and improved materials are needed. Bone marrow aspirate (BMA) is the most practical source of multipotent stromal cells and some studies have reported enhanced bony ingrowth within hydroxyapatite (HA) scaffolds. This study investigated the effect of the binding agent Calcium/Sodium Alginate fibre gel and the addition of autogenic BMA on bone growth into a porous HA scaffold manufactured using novel patented techniques and implanted in an ovine femoral condyle critical-sized defect. Our hypothesis was that Alginate fibre gel would have no negative effect on bone formation and osteoconduction within the scaffold and that BMA would augment the bone turnover rate and incorporation of the graft with the surrounding bone at 6 and 12 weeks post implantation.

Comparison of Proliferation and Osteoblast Differentiation of Marrow-Derived Mesenchymal Stem Cells on Nano- and Micro-Hydroxyapatite Contained Composite Scaffolds
Bones constructed by tissue engineering are being considered as valuable materials to be used for regeneration of large defects in natural bone. In an attempt to prepare a new bone construct, in this study, proliferation and bone differentiation of marrow-derived mesenchymal stem cells (MSCs) on our recently developed composite scaffolds of nano-, micro-hydroxyapatite/ poly(l-lactic acid) were ...
متن کاملComparative Study of Bone Repair Using Porous Hydroxyapatite/ β-Tricalcium Phosphate and Xenograft Scaffold in Rabbits with Tibia Defect
Background: Bone tissue engineering requires materials that are biocompatible, mechanically suited for bone function, integrated with the host skeleton, and support osteoinduction of the implanted cells for new bone formation. The aim of this study was to compare the osteogenic potential of xenograft with hydroxyapatite/β- tricalcium phosphate (HA/β-TCP) scaffold. Methods: New Zealand rabbits (...

Background. This work compares the osteoblastic behaviour of a bone marrow (BM) aspirate and a prepared BM concentrate of nucleated cells associated with a glass reinforced hydroxyapatite composite (GRHC) in a microporous pellet formulation. Methods. BM aspirate (30 mL) was collected during 3 orthopedic surgical procedures, and a concentration system was used to achieve 3 rapid preparations of ...
متن کاملImproving the mechanical and bioactivity of hydroxyapatite porous scaffold ceramic with diopside/forstrite ceramic coating
Objective(s): Scaffolds are considered as biological substitutes in bone defects which improve and accelerate the healing process of surrounding tissue. In recent years a major challenge in biomaterials is to produce porous materials with properties similar to bone tissue. In this study, the natural bioactive hydroxyapatite scaffolds with nano Diopside /Forstrite coating was successfully synthe...
متن کاملThe Augmentation of a Collagen/Glycosaminoglycan Biphasic Osteochondral Scaffold with Platelet-Rich Plasma and Concentrated Bone Marrow Aspirate for Osteochondral Defect Repair in Sheep
Objective: This study investigates the combination of platelet-rich plasma (PRP) or concentrated bone marrow aspirate (CBMA) with a biphasic collagen/glycosaminoglycan (GAG) osteochondral scaffold for the treatment of osteochondral defects in sheep. Design: Acute osteochondral defects were created in the medial femoral condyle (MFC) and the lateral trochlea sulcus (LTS) of 24 sheep (n = 6). Def...
